Cover Story

The cover image shows a monkey kidney vero cell: the two nuclei are stained with DAPI (blue); the vesicles with yellow fluorescent protein (green); and the mitochondria with mitotracker (red).

Olympus has introduced its latest and most advanced range of highly versatile, cutting-edge fluorescence research microscope systems offering motorisation, user control and a unique level of versatility for life sciences. The xcellence line provides the ultimate technology for the latest live cell techniques, including real-time, high-speed imaging, TIRFM and HILO (cell^tirf), and photocontrol such as FRAP, FLIP and photoactivation (cell^frap). All Olympus xcellence systems are designed to bring the most advanced fluorescence imaging techniques within easy reach of the user. Experiment Manager  provides a simple drag and drop method, designed for streamlining even the most complex and interactive experiments. Advanced fluorescence light sources and very fast filter, attenuator and shutter changes enable the capture of clearly resolved images.

The cell^tirf module enables ultra-sensitive, simultaneous multicolour total internal reflection fluorescence (TIRF) microscopy. It features four laser lines with separate motorised angle control for synchronised acquisition with identical penetration depth. Olympus has introduced a peerless range of TIRF objectives, including the highest magnification currently on the market – 150 x and the highest NA of 1.65, enabling highly precise investigations to be carried-out at penetration depths down to 40 nm.

The cell^frap system offers easy and fast real-time photobleaching as well as photomanipulation, including FRAP, iFRAP, FLIP, FLAP, photoactivation and photoswitching. The speed and precision of the cell^frap module is made possible through the combination of the real-time controller and CCD camera-based imaging, which is significantly faster than point scanning confocal-based FRAP.
